Catalog Number: (PRSI33-476)
Supplier: ProSci Inc.
Description: Recognises a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This mAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyses several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this mAb shows tyrosinase in melanotic as well as amelanotic variants. This mAb is a useful marker for melanocytes and melanomas.

Description: AFP Antibody: Alpha-fetoprotein (AFP) is a major plasma protein produced by the yolk sac and the liver during fetal development. In adults, AFP expression is often associated with hepatoma or teratoma. However, hereditary persistance of AFP may also be found in individuals with no obvious pathology. AFP is thought to be the fetal counterpart of serum albumin, and the AFP and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. AFP is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. AFP levels in amniotic fluid is used to measure renal loss of protein to screen for spina bifida and anencephaly.

Description: Superoxide Dismutase [Cu-Zn] (SOD1) is a soluble cytoplasmic and mitochondrial intermembrane space protein that belongs to the Cu-Zn superoxide dismutase family. SOD1 binds copper and zinc ions and is one of three isozymes responsible for destroying free superoxide radicals in the body. SOD1 neutralizes supercharged oxygen molecules, which can damage cells if their levels are not controlled. The enzyme protects the cell against dangerous levels of superoxide. Zinc binding promotes dimerization and stabilizes the native form. Mutations in SOD1 cause a form of familial amyotrophic lateral sclerosis. Defects in SOD1 are the cause of amyotrophic lateral sclerosis type 1 (ALS1) which is a familial form of amyotrophic lateral sclerosis, a neurodegenerative disorder affecting upper and lower motor neurons and resulting in fatal paralysis.

Description: STEAP1 Antibody: The six-transmembrane epithelial antigen of prostate 1 (STEAP1) was the first member of a family of metalloreductases identified as cell-surface antigens in prostate tissue. The normal function of STEAP is still uncertain; unlike other members of the STEAP family, STEAP1 does not promote iron or copper reduction or uptake and lacks the FNO-like reductase domain critical for activity. However, its expression is highly increased in multiple cancer cell lines, including prostate, bladder, colon, and ovarian cancers. Supporting this is evidence that STEAP1 peptides can be used to stimulate CD8+ T cells from healthy donors, enabling them to recognize STEAP1-positive human tumor cells, suggesting that STEAP1 may a potential target for cancer immunotherapy. At least three isoforms of STEAP1 are known to exist.

Supplier: WTW
Description: A wide choice of tests is available, for routine quantitative analysis in different applications, designed for use with WTW spectrophotometers and photometers. The test is automatically recognised by the laboratory instruments (via a barcoding system) and results are displayed in the appropriate units. Due to the different photometer optics, one test may yield different measuring ranges in different instruments; LED photometers may have smaller measuring ranges for the same test.

Catalog Number: (BOSSBS-0225M)
Supplier: Bioss
Description: Keyhole limpet hemocyanin is an extremely large, heterogeneous glycosylated protein consisting of subunits with a molecular weight of 350,000 and 390,000 in aggregates with molecular weights of 4,500,000-13,000,000. Each domain of a KLH subunit contains two copper atoms that together bind a single oxygen molecule (O2). When oxygen is bound to hemocyanin, the molecule takes on a distinctive transparent, opalescent blue color. The KLH protein is potently immunogenic yet safe in humans and is therefore highly prized as a vaccine carrier protein. The large and highly glycosylated KLH protein cannot be reproduced synthetically. It is available only as a purified biological product from the Keyhole Limpet Megathura crenulata.Keyhole limpet hemocyanin (KLH) is used extensively as a carrier protein in the production of for research, biotechnology and therapeutic applications. Haptens are substances with a low molecular weight such as peptides, small proteins and drug molecules that are generally not immunogenic and require the aid of a carrier protein to stimulate a response from the immune system in the form of antibody production.[2] KLH is the most widely employed carrier proteins for this purpose. KLH is an effective carrier protein for several reasons. Its large size and numerous epitopes generate a substantial immune response, and abundance of lysine residues for coupling haptens, allows a high hapten:carrier protein ratio increasing the likelihood of generating hapten-specific . In addition, because KLH is derived from the limpet, a gastropod, it is phylogenetically distant from mammalian proteins, thus reducing false positives in immunologically based research techniques in mammalian model organisms.KLH may also be a challenging molecule to work with because of its propensity to aggregate and precipitate. Aggregates remain immunogenic, but limit the ability to conjugate haptens and are difficult to manipulate in the laboratory.

Description: This gene encodes a member of the lysyl oxidase gene family. The prototypic member of the family is essential to the biogenesis of connective tissue, encoding an extracellular copper-dependent amine oxidase that catalyses the first step in the formation of crosslinks in collagens and elastin. A highly conserved amino acid sequence at the C-terminus end appears to be sufficient for amine oxidase activity, suggesting that each family member may retain this function. The N-terminus is poorly conserved and may impart additional roles in developmental regulation, senescence, tumor suppression, cell growth control, and chemotaxis to each member of the family. Alternatively spliced transcript variants of this gene have been reported but their full-length nature has not been determined.

Description: SCO2 Antibody: Synthesis of cytochrome c oxidase 2 was initially identified in yeast as one of two cytochrome c oxidase (COX) assembly proteins that enable the assembly of cytochrome c holoenzyme, a complex that catalyzes the transfer of reducing equivalents from cytochrome c to molecular oxygen and pumps protons across the inner mitochondrial membrane. Like their yeast homologs, the function of both SCO2 and SCO1 are dependent on copper ion binding. Recent studies suggest that SCO2 expression is regulated by p53, so that a decrease in p53 expression, such as in numerous tumors and cells lines, the drop in SCO2 expression leads to a shift from normal aerobic respiration towards the production of glycolytic ATP. Defects in the SCO2 protein are also associated with fatal infantile cardioencephalomyopathy and COX deficiency.
