Description: FANCA (also called Protein FACA or Fanconi anemia group A protein) is involved in DNA repair, perhaps specifically with post-replication repair or a cell cycle checkpoint function. FANCA may also be implicated in interstrand DNA cross-link repair and in the maintenance of normal chromosome stability. The Fanconi anemia complementation group (FANC) currently includes FANCA, FANCB, FANCC, FANCD1 (also called BRCA2), FANCD2, FANCE, FANCF, FANCG, and FANCL. The previously defined group FANCH is the same as FANCA. Fanconi anemia is a genetically heterogeneous recessive disorder characterized by cytogenetic instability, hypersensitivity to DNA crosslinking agents, increased chromosomal breakage, and defective DNA repair. The members of the Fanconi anemia complementation group do not share sequence similarity; they are related by their assembly into a common nuclear protein complex. This gene encodes the protein for complementation group A. Alternative splicing results in multiple transcript variants encoding different isoforms. Variant 1 (isoform a) encodes the longest transcript. Variant 2 (isoform b) contains an alternate exon, which results in an early stop codon, compared to variant 1. Description: HEPACAM2 Antibody: HEPACAM2 (Hepatocyte cell adhesion molecule 2), a type I N-linked transmembrane glycoprotein, belongs to the immunoglobulin superfamily. The exact function of HEPACAM2 is currently unknown, but the related protein HEPACAM forms cishomodimers on the cell surface to regulate the cell adhesion and may inhibit cell growth through suppression of cell proliferation. HEPACAM and HEPACAM2 mRNA are differentially regulated in canine tumors, with HEPACAM2 mRNA showing increased levels in adenomas, but decreased levels in metastatic carcinomas compared to normal tissues, while HEPACAM protein levels decreased in adenomas. It is therefore likely that HEPACAM2 plays a different role than HEPACAM in the development and progression to tumors.

When compared to LAIR-1, its transmembrane counterpart, it shares 83% amino acid identity across the signal sequence and extracellular domains; although one is secreted and one is membrane-bound, the two LAIR proteins are thought to have arisen from a common gene ancestor and appear to share similar adhesion profiles. This suggests that LAIR-2 may compete with LAIR-1 for ligand binding. A 114 amino acid alternate splice form of LAIR-2 is truncated at the C terminus, but retains the entire Ig domain. The expression profile of these splice forms, and the presence of orthologs in other species, have not been reported.

This gene product was shown to bind single-stranded DNA ends, and mediate the DNA-DNA interaction necessary for the annealing of complementary DNA strands. It was also found to interact with DNA recombination protein RAD51, which suggested its role in RAD51 related DNA recombination and repair. Four alternatively spliced transcript variants encoding different isoforms have been reported for this gene. The alpha variant encodes the longest isoform that contains an identical N-terminus, but a distinct C-terminus, as compared to other isoforms (beta, delta, and gamma).
