Description: CYP51A1 is a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyse many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This endoplasmic reticulum protein participates in the synthesis of cholesterol by catalysing the removal of the 14alpha-methyl group from lanosterol. Homologous genes are found in all three eukaryotic phyla, fungi, plants, and animals, suggesting that this is one of the oldest cytochrome P450 genes. Two transcript variants encoding different isoforms have been found for this gene.

Description: This gene encodes a member of the cytochrome P450 superfamily of enzymes. The cytochrome P450 proteins are monooxygenases which catalyze many reactions involved in drug metabolism and synthesis of cholesterol, steroids and other lipids. This protein localizes to the mitochondrial inner membrane and catalyzes the conversion of cholesterol to pregnenolone, the first and rate-limiting step in the synthesis of the steroid hormones. Two transcript variants encoding different isoforms have been found for this gene. The cellular location of the smaller isoform is unclear since it lacks the mitochondrial-targeting transit peptide. Description: High density lipoproteins (HDLs) have been proposed to function jointly with lecithin:cholesterol acyltransferase and CETP to facilitate cholesterol transport from tissues to the liver. This mechanism, referred to as reverse cholesterol transport, is physiologically important because it maintains systemic cholesterol levels. CETP is responsible for neutral lipid transfer activity in plasma in numerous species. Since CETP is able to accelerate specifically the exchange of lipid components between pro- and anti-atherogenic lipoprotein fractions, it may be a key determinant of the global atherogenicity of the plasma lipoprotein profile and arises as a possible target in atherosclerosis prevention. CETP has an important role in reverse cholesterol transport and shaping and affecting the composition of plasma lipoproteins. In general elevated levels of CETP have been associated with increased risk of coronary heart disease.

Description: LDL and HDL transport both dietary and endogenous cholesterol in the plasma. LDL is the main transporter of cholesterol and cholesteryl esters and makes up more than half of the total lipoprotein in plasma. LDL is absorbed by the liver and other tissues via receptor mediated endocytosis. The cytoplasmic domain of the LDL receptor facilitates the formation of coated pits; receptor-rich regions of the membrane. The ligand binding domain of the receptor Recognises apo-B100 on LDL, resulting in the formation of a clathrin-coated vesicle. ATP-dependent proton pumps lower the pH inside the vesicle resulting dissociation of LDL from its receptor. After loss of the clathrin coat the vesicles fuse with lysozomes, resulting in peptide and cholesteryl ester enzymatic hydrolysis. The LDL receptor can be recycled to the cell membrane. Insulin, tri-iodothyronine and dexamethasome have shown to be involved with the regulation of LDL receptor mediated uptake. The protein component of LDL is apolipoprotein B100. LDL contains 2022% protein, 1015% triglycerides, 2028% phospholipids, 3748% cholesteryl esters and 810% cholesterol.

Description: Apolipoproteins are protein components of plasma lipoproteins. ApoM (Apolipoprotein M), also known as protein G3a, is a member of the Lipocalin family of proteins. ApoM is exclusively expressed in kidney tubular epithelial cells and liver hepatocytes. Mature ApoM retains its signal peptide, which acts as a hydrophobic anchor, and contains a structurally conserved eight stranded antiparallel ∫ barrel which binds retinol and retinoic acid. ApoM may play a key role in reverse cholesterol transport. It mainly associates with high density lipoprotein (HDL) and to a lesser extent with triglyceride-rich lipoprotein (TGRLP) and low-density lipoprotein (LDL). ApoM is important for the pre∫-HDL formation. Pre∫-HDL is an important acceptor of peripheral cellular cholesterol. The concentration of ApoM in plasma strongly correlates with total cholesterol. Low concentrations of ApoM in plasma is associated with diabetes.
