Sulphamethoxazole

Sulphonamide antibiotic that blocks the synthesis of dihydrofolic acid by inhibiting the enzyme dihydropteroate synthase. Mode of Action: Inhibits folic acid synthesis in prokaryotes. Anti-microbial Spectrum: Gram positive, Gram negative, Chlamydia. Mode of Resistance: Alteration of dihydropteroate synthase or alternative pathway for folic acid synthesis.


Sulphamethoxazole is a member of the sulphonamide family of antibiotics. sulphamethoxazole interferes with folic acid synthesis in susceptible bacteria. sulphamethoxazole and other sulphonamides have a similar structure to p-aminobenzoic acid. These compounds interfere with the synthesis of nucleic acids in sensitive microorganisms by blocking the conversion of p-aminobenzoic acid to the co-enzyme dihydrofolic acid, a reduced form of folic acid.


Slphamethoxazole is used for treating various types of bacterial infections, including urinary tract infections, acute worsening of chronic bronchitis, and ear infections. In addition it can be used as an alternative to amoxicillin-based antibiotics to treat sinusitis.


Practically insoluble in water, freely soluble in acetone, sparingly soluble in ethanol. It dissolves in dilute solutions of sodium hydroxide and in dilute acids.
